Blood clotting enzyme Essay Serine protease proteins are important enzymes involved in the process of blood coagulation. Blood coagulation is an importance defense mechanism that prevents the host mammal organism from losing excess blood or from forming unwanted blood clot. The process of coagulation can be initiated by both intrinsic factors and extrinsic factors. A cascade of event is followed which activate these enzymes; normally the enzymes are inactive state a condition called zymogens. Zymogens by their virtual condition of being inactive prevent unwanted blood clotting which may have a far reaching consequence such as thrombosis. Blood clotting in a series of processes, in which the zymogensââ¬â¢ need to be activated by reacting with its glycoprotein co-factors. Among the serine protease is the thrombin enzyme factor five (v) responsible for clearing clot in the blood. The enzyme is usually present circulating in plasma which is made up of a single monomer chain, it life span can range from 12 to 36 hours. In human the main regulator in blood coagulation is erythrocytes leukemia cells which activate adenylate cyclase, the process is reversible by the interaction of Aalpa-thrombin with glycoprotein while b alpha enhance the platelets interaction which initiate the proteolytic process. Fibrin-bound thrombin is cleaved by thrombin at a very specific site at the extracellular N-terminal, PAR-1 regulates a number of endothelial cell biology, vascular development but more so is a mediator of thrombin signaling. The human thrombin consist of two gamma chains namely the gamma A and gammaââ¬â¢ the final stage of coagulation of mammalian blood involves the cleavage of the four arginine and glycine bond. Binding studies shows both fibrin 1 and fibrin 2 with low affinity to the E domain and high binding affinity at the extreme end of 408 to 425 on the gamma chain The mode of action involves conversion of fibrinogen to fibrin by breaking the bonds in the fibrinogen at a precise position of arginine and glycine where the fibrin peptides are released. The serine proteases require restructuring itself in order to fit the key and lock model. The glycine at the position five is highly conserved because it is the one which occupy the active site which is determine by the acryl group during the substrate conformation. A study done using heparin indicated an overlap of the active site, which was attributed to the interaction of thrombin and the gamma peptide chains to the external and the interaction to the active site in close proximity to the Na+ of the substrate. The role of the enzyme can be explained in terms of NA+ binding to thrombin on the basis of prothrombotic and procoagulant. The cascade is a continuous cycle of events that are activated by two factors ,factor (ix) and factor (viii) to form tenase complex which is discontinued by down regulation that occur in the following mechanism which include; serpin (serine protease inhibitors) which function to degrade thrombin and other activation factors, it can also be regulated by protein C where thromodulin bind to it and is inactivate in the presence of protein S, limiting the action of tissue factor by the tissue factor pathway inhibitors by inhibiting excessive TF mediated activation of factor (ix) and factor (x), plasmin help to degrade fibrin hence preventing more fibrin being formed and lastly but not the least regulation through adenylate cyclase pathway by inhibiting platelet activation by decreasing cytosol level of calcium which ultimately result to decreased release of granules which are responsible for the activation of more platelets and coagulation cascade. In conclusion the work play by thrombin enzyme is of paramount importance considering the complexity of the enzyme kinetics involved in the human body. The writer choose Margareth Thatcher as the object of the writers study because the writer wants to reveal that in the past time even in recent days women are oppressed in a political field, they have not been given chances to show their ability to lead the society. However, Margareth Thatcher shows her ability to carry out the Britains government. Margareth Thatcher was born on October 13, 1925, in Grantham, England. She is the daughter of a local businessman who was introduced to conservative politics by her father, a member of the towns council. When she was in university, she served as a president of the Conservative Association. Two years after she graduated from her college, she accepted to work for public office. She ran as the conservative candidate for a Dart ford parliamentary seat in the 1950 elections. She was elected to be a leader of the Conservative Party in 1975. She served as the prime minister of England from 1979 to 1990. As a prime minister, she battled the countrys recession by initially raising interest rates to control inflation. She can run the country quite well marked with some progression that has been occurred in that time. However, when we are speaking about gender roles in such a stratified society, the class struggle is simply a cause to gender stereotypes and exploitation. (Madsen, 2000) Gender stereotypes is actually a set of values of being proper and improper to be done by a woman which is functioned as a limitation These values often create an unfriendly environment or even such condition for women to take part in legal and social functions. Furthermore, it supports a condition that womens rights are susceptible to violation. There are many kinds of stereotypes and several reasons of stereotypes. Sex stereotypes, are supposedly based on the physical or biological differences. Sexual stereotypes are those based on the perceived characteristics that play a role in sexuality, including, for example, sexual initiation, intimacy, possession, and objectification while sex role stereotypes describes a normative or statistical view regarding appropriate roles or behavior for men and women. Josephson (2005) in her book Gender and American Politics: Women, Men, and the Political Process: Tolleson-Rinehart explains the examples of discrimination among women and men government workers in western legal system. She says that among others, women usually suffer in terms of peoples trust, media coverage, and self-inconfidence from their male counterparts. Based on Gelbs theory of feminism and politics: a comparative perspective (1989), there are two kinds of feminist movement in power-seizing political fields, they are the reformists and radicals. Both of them move based on their own ideology of how women should be participating in such conditions. According to Jonasdottir (1994), oppression based on gender stratification is similar to Marxist class division where the gender woman has been named as the second gender. Thus it makes women are vulnerable to discriminate based on what Marxism defines as materialistic and economic value. The conceptual tools of cinematography Blain Brown (2011, p.3) in his book Cinematography Theory and Practice: Image Making for Cinematographers and Director says that the tools of cinematography and filmmaking actually discuss the essential techniques, methods, and elements that we can use to tell the story. Some elements in cinematography visualization which can be used to support this study are camera angles, shot sizes, and techniques of framing. According to Bernard Dick, a shot is simply what is recorded by a single operation of the camera. He also states that it can also be defined in terms of distance. (1998, pp.36-37). In other words, shot is a matter of subjects size or amount in the frame. It embodies five types; they are close up (CU), extreme close up (ECU), long shot (LS), extreme long shot (ELS), and medium shot (MS). He also says that shots are also defined by the position of the camera in relation to the subject or what do we known as angle. It is determined by where the camera is placed. There are four types of angle; they are birds eye view, high-angle (h/a), low-angle (l/a), and oblique-angle. In framing techniques, there is a rule called rule of thirds. Rule of thirds is a guideline which applies to the process of composing visual images such as paintings, photographs, and designs. It proposes that an image should be imagined as divided into nine equal parts by two equally-spaced horizontal lines and two equally-spaced vertical lines, and that important compositional elements should be placed along these lines or their intersections. When we are photographing or filming people, it is common to line the body up with a vertical line, and having the persons eyes in line with a horizontal one. (2012, Rule of Thirds, à ¶.1). A Most Remarkable Teacher à A most remarkable high school teacher first aroused my interest in medicine when he invited me to take a special advanced microbiology course at night. He (Mr. Wiles) had worked for years in the medical field before retiring to teach school. Spending many hours with us, he constantly reaffirmed his belief that we could make a positive contribution to society through a career in medicine. Shortly after my graduation, Mr. Wiles died of lung cancer. During his last months, as I visited him, walked with him, chopped wood for him, I anguished over my helplessness as his pain grew and his life diminished. The doctors couldn't do much to reverse that process, but at least they could t~ to ease his pain or Blow the spreading disease. I admired and envied that ability. à In the ensuing years, I found myself embracing two seemingly different career paths. I loved being around people and thrived on the interaction, but I was fascinated by the technical intricacies of science. With a love for my science courses, I found that I could readily follow the logic of scientific thought. Then I left school and went to Europe as a volunteer missionary. In addition to learning a foreign Language (French) â⬠¢ I spent that year and a half learning to work with people and help them with their problems. This strengthened my desire to work with people in some kind of clinical setting. à It was... ...e good, some not as good--as they made their rounds. The really good ones seemed able to comfort their patients in more than just a physical sense. One oncologist seemed never to fail to bring a smile to his patients' faces, such was his genuine caring. à I believe, now, the words of Mr. Wiles. I believe that as a physician I can make a very real contribution to society. I can see myself offering all I have to help a patient smile again, using all my resources to find the best scientific solution. And as I become more of a scientist, so must I become even more skilled with people--people with needs and feelings. For medicine is the culmination of that art and science-requiring the best of both worlds.

Air Accident An Air accident is the worst nightmare of every pilot or passenger that has ever ridden in an aircraft. Although air travel is one of the safest forms of transportations, accidents do happen with dramatic and terrifying results. The circumstance of Air accidents takes place by four causes: takeoff and landing, mechanical failures, pilot error, and bad weather. One cause of Air accidents take place is during takeoff and landing. Approximately 80 percent of all aviation accidents occur shortly before, after, or during takeoff or landing, and are often described as resulting from ââ¬Å"human error. The causes of Air accident is vary greatly depending on problems that may develop during the flight possess. In many situations these accidents can be completely avoided through careful preparation and effective safety techniques. Taxi and Takeoff is one of the most important parts of a flight. The preflight and planning stages of the operation must be carefully done to prevent accidents and save many lives. Landing Accidents during Descent account for 36 percent of all general Air mishaps and are the most common type of accident.When combined, taxi and takeoff accidents account for about 22percent jet airplane accidents, and about 22percent of all fatalities. Another cause of Air accidents is mechanical failures. No form of transportation is completely safe, and no machine is completely foolproof. Mechanical failure is responsible for 13percent of all commercial airplane accidents. An airplane can crash if engine failure can come in form of simple stalling. It could be as worse as a complete detachment of the motor itself.Many Modern planes have more than one engine and this allows the plane to continue to fly when one is shut down. However, these other cases where all shut at one, or one after the other, and accident occur. Anything that can go wrong will go wrong. Tragically, this sometimes applies to Air accidents when mechanical failures occur. A some what different cause for Air accidents is a pilot error. When most people make mistakes at their jobs, their employment can be terminated.In contrast, when pilots are negligent or make errors while on their jobs, there is potential for hundreds of lives to be terminated. The pilots receive extensive training designed to prepare them to handle a wide variety of situations, but there are times when fatal mistakes are made. When flight crew and pilots do their jobs correctly, Air accidents are much less likely to occur. Another important cause of Air accidents is bad weather. For example, flying at peak thunderstorm times should be avoided.In addition, flying at night is no safer than flying in the day. The weather caused 4percent of Air accidents. However, other problems made up the remaining 4percent. Most plane crashes happen in the months of September and December. However, no one from any investigation knows almost Air accidents happened in these two months. In conclusion, the cir cumstance of Air accidents takes place by four causes: takeoff and landing, mechanical failures, pilot errors, and bad weather. Yet many airplane accidents still have unknown causes.
